New Technique Makes Early Human Embryos From Skin Cells
Researchers have created human skin-cell-derived eggs that can form early human embryos by combining cloning technology, fertilization and simple chemical methods, according to a new study published in Nature Communications.
Scientists have successfully produced reproductive cells of many animals, like pandas, yet creating human eggs and sperm has long been a tough task. This new technology is expected to treat infertility for women affected by aging, early menopause or cancer treatments. It may also enable same-sex male couples to have biologically related children, experts suggest.
The research adopted the somatic cell nuclear transfer technology used for cloning. Researchers replaced the nucleus of a human egg with that of a human skin cell. Different from common body cells with 46 chromosomes, mature eggs need only 23 chromosomes through meiosis. When fertilized by sperm, normal eggs can form embryos with 46 chromosomes.
However, cloned human eggs failed to remove half of their chromosomes after fertilization. To solve this problem, researchers used a chemical molecule to help cut chromosome numbers. Some fertilized eggs grew into early embryos, but most failed due to abnormal chromosome numbers. The embryos only developed for about six days and none grew into viable ones, with wrong chromosome counts and missing or repeated chromosomes.
Scientists explained the failure lay in random chromosome pairing during cell division. Though imperfect, the study proves the feasibility of this technology. Researchers estimate it will take at least ten years for clinical tests. Besides, such tests are banned in the US for genetic modification rules.
Experts point out the technique’s major shortcoming is its need for donor eggs. Even so, it marks a key breakthrough in human gene research and will inspire more new technologies in the future.
